Somdev Devvarman's lost to 23rd seed Philipp Kohlschreiber of Germany in the second round of the US Open.
Kohlschreiber proved too much experienced for the Indian and won 6-3, 6-4, 0-6, 6-2.
Somdev had won three qualifying matches to make it into the main draw at the tournament. He won his first-round match against Frederico Gil of Portugal in straight sets.
Sania Mirza, meanwhile, overcame from her singles defeat to emerge victorious in the mixed doubles and women's doubles events.
Sania and Francesca Schiavone of Italy beat Ekaterina Dzehalevich and Renata Voracova 5-7, 7-5, 6-1 to enter the second round of the women's doubles. They will meet Gisela Dulko and Shahar Peer.
In the mixed doubles, Sania Mirza and Canada's Daniel Nestor beat Akgul Amanmuradova and Ashley Fisher 6-3, 6-1 to enter the second round.
On Wednesday, Sania's singles campaign wilted in the second round with 10th seeded Italian Flavia Pennetta routing the Indian 6-0, 6-0 in a grossly lop-sided match.
India's Leander Paes and his Czech partner Lukas Dlouhy have already moved into the second round of the men's doubles event with a win over Romanian pair of Victor Hanescue and Grabriel Trifu in first round.
